苏淮猪SULT1C3基因多态性的检测与发情期血浆中雌激素水平的测定

本研究针对SULT1C3基因位点进行了引物设计,并以提取的42头苏淮猪核心群母猪的DNA为模板,进行了PCR扩增反应,对扩增产物进行DNA测序,得到了测序结果。

摘要:SULT1C3具有硫酸基转移酶活性和醇磺基转移酶活性,在雌激素的降解过程中发挥关键性作用。发情期外周血雌激素水平决定母猪发情征状,而发情征状的不明显会导致母猪配种率的降低,最终影响猪场的繁殖效率。本研究以苏淮猪为对象,测定发情期苏淮猪雌激素水平,并在对SULT1C3进行多态性检测的基础上,分析了二者之间的相关性,旨在筛选出与苏淮猪雌激素水平显著相关的多态位点,为改善苏淮猪繁殖性能奠定基础。方法:采集发情期苏淮母猪的耳组织和血液样品,通过放射免疫法测定血清中雌激素和孕酮浓度,对SULT1C3基因型进行测序,并利用单因素方差分析血清雌激素水平与母猪SULT1C3基因型之间的关系。结果:存在突变位点,但不同基因型之间雌激素水平差异不显著(P>0.05)。

Polymorphism of SULT1C3 gene and plasma estrogen concentration of Suhuai pigs at estrus

Abstract:SULT1C3 has the activity of sulfuric acid transferase and alcohol sulfotransferase, which plays a critical role in the degradation of estrogen. Estrogen levels in the estrus period determine the estrus symptoms. Estrus not obvious may lead to a decrease of sows breeding rate, and ultimately affect the reproductive efficiency of pig. In this study, the estrogen level of Suhuai pig was measured. Based on the polymorphism detection of SULT1C3, the correlation between the two was analyzed, in order to screen out the polymorphic loci that were significantly related to estrogen levels in Suhuai pigs. The results will lay the foundation for improving the reproductive performance of Suhuai pig. Methods: The ear tissue and blood samples of Suhuai sows were collected. The estrogen and progesterone in the serum were measured by radioimmunoassay. The SULT1C3 genotype was sequenced and the estrogen level was compared with the sow SULT1C3 genotype. Results: There were no significant differences in estrogen levels among different genotypes (P> 0.05).
